"Can you cast the spell more than once on the same camp site or room such that intruders would have to roll multiple saving throws?"

Nothing prevents it in the description. However, IMHO, for this to work it likely requires each casting to have a different "center point" (as it uses radius of effect, overlapping coverage is fine).
